چکیده

South Africa plays a critical international role in white rhino, Cerathotherium simum conservation. The Mpumalanga Parks Board (MPB) is the statutory organization entrusted with conservation in the Province. The MPB hosts key populations of white rhino on some of its protected areas. None of these populations is currently or potentially large enough to be viable on its own. Challenges for an expansion in numbers are the limited habitat available and intra-specific competition. A meta-population strategy is required to overcome these challenges and to prevent inbreeding. The effective management of small sub-populations within a meta-population strategy requires the identification and permanent recognition of individual rhino. This has been achieved in the recent past through micro-chipping, ear-notching and the compilation of an individual file for each animal. Despite the intensive documentation and monitoring of these sub-populations, the extent to which individual bulls are responsible for successive generations of calves cannot be established with certainty. Genetic data are required. A DNA profiling technique based on molecular genetics was developed by the Animal Improvement Institute of the Agricultural Research Council using material collected from the MPB rhino. Individual animals can now be unequivocally identified by means of unique bands of micro-satellites. Each individual profile consists of a combination of the two parent’s profiles, making it therefore possible to construct parental lineages. A genetic database of the MPB rhino is being constructed. The information from the database is being applied to determine parentage, animal productivity, genetic variation, rate of inbreeding and genetic distances between populations. The DNA profiling technique provides a powerful tool for the MPB to preventing deleterious genetic consequences of maintaining small, isolated sub-populations. Objective decisions can now be made as to which individual rhino should be exchanged amongst the different sub-populations. 0199_inter_onc_tome2_part7 19/10/05 20:25 Page 787

The Sixth Rhino: A Taxonomic Re-Assessment of the Critically Endangered Northern White Rhinoceros

BACKGROUND The two forms of white rhinoceros; northern and southern, have had contrasting conservation histories. The Northern form, once fairly numerous is now critically endangered, while the southern form has recovered from a few individuals to a population of a few thousand. Since their last taxonomic assessment over three decades ago, new material and analytical techniques have become avai...

Social and spatial relationships in captive southern white rhinoceros (Ceratotherium simum simum).

Although critical to the conservation of white rhinoceros, captive breeding has proven challenging because of the poor and irregular reproductive health of many captive rhinos, and social interactions may play a significant role. This research investigated the social and spatial relationships of two captive groups of southern white rhinoceros (Ceratotherium simum simum) by examining the frequen...

Effective Use of Synthetic Seed Technology in the Regeneration of Dendrobium White Fairy Orchid

The synthetic seed technology is becoming popular due to its wide application in germplasm conservation and for exchanges between countries in the floriculture trade. In this study, this method was used to study the germination and conversion capabilities of orchid species Dendrobium White Fairy when stored at different storage times and with different storage containers. A high germination per...

SNP discovery and characterisation in White Rhino (Ceratotherium simum) with application to parentage assignment

The white rhino is one of the great success stories of modern wildlife conservation, growing from as few as 50-100 animals in the 1880s, to approximately 20,000 white rhinoceros remaining today. However, illegal trade in conservational rhinoceros horns is adding constant pressure on remaining populations. Captive management of ex situ populations of endangered species using molecular methods ca...

Strategies and Clinical Applications of Next Generation Sequencing

Abstract DNA sequencing is one of the great valuable techniques in molecular biology, which can be used to detect the sequence of nucleotides in a DNA fragment. The high-throughput sequencing known as Next Generation Sequencing (NGS) revolutionized genomic research and molecular biology; therefore, the whole human genome can be sequenced with a low cost in several days. NGS technology is simi...
